    Detecting variable responses in time-series using repeated measures ANOVA: Application to physiologic challenges.

    We present an approach to analyzing physiologic timetrends recorded during a stimulus by comparing means at each time point using repeated measures analysis of variance (RMANOVA). The approach allows temporal patterns to be examined without an a priori model of expected timing or pattern of response. The approach was originally applied to signals recorded from functional magnetic resonance imaging (fMRI) volumes-of-interest (VOI) during a physiologic challenge, but we have used the same technique to analyze continuous recordings of other physiological signals such as heart rate, breathing rate, and pulse oximetry. For fMRI, the method serves as a complement to whole-brain voxel-based analyses, and is useful for detecting complex responses within pre-determined brain regions, or as a post-hoc analysis of regions of interest identified by whole-brain assessments. We illustrate an implementation of the technique in the statistical software packages R and SAS. VOI timetrends are extracted from conventionally preprocessed fMRI images. A timetrend of average signal intensity across the VOI during the scanning period is calculated for each subject. The values are scaled relative to baseline periods, and time points are binned. In SAS, the procedure PROC MIXED implements the RMANOVA in a single step. In R, we present one option for implementing RMANOVA with the mixed model function "lme". Model diagnostics, and predicted means and differences are best performed with additional libraries and commands in R; we present one example. The ensuing results allow determination of significant overall effects, and time-point specific within- and between-group responses relative to baseline. We illustrate the technique using fMRI data from two groups of subjects who underwent a respiratory challenge. RMANOVA allows insight into the timing of responses and response differences between groups, and so is suited to physiologic testing paradigms eliciting complex response patterns

    Religion and urban regeneration: a place for faith?

    The British government has identified 'faith communities' as a neglected resource in urban regeneration. This article first explores the context of official support for faith involvement in urban and neighbourhood policy and identifies the assumptions underlying key policy documents. These assumptions are then critically explored by reference to the links commonly drawn between religion and 'community', 'neighbourhood' and 'social cohesion'. Attempts to enlist faith groups within this essentially consensual agenda often fail to recognise both the potential divisiveness of religion and also the more positive and radical lessons that often stem from the action, experience and critical analysis of religious organisations and their members.</p

    Student-centered interventions the key to student health care worker influenza vaccination

    Objectives: To investigate influenza vaccination uptake rates, attitudes and motivations towards influenza vaccination among student health care workers (HCWs). Methods: Self-reported influenza vaccination uptake among student HCWs at The University of Notre Dame Australia, Fremantle, Western Australia (UNDAF) was surveyed before and after implementation of a peer-led, student-centered campaign to raise awareness of, and improve access to, influenza vaccination. Data were weighted and analysed using logistic regression. Results: Pre-campaign influenza vaccination uptake was 36.3% (95% CI=31.8%-40.8%), with students identifying lack of awareness of both the Australian Government’s recommendations and university policy, cost, and inconvenience of vaccine access as key barriers. Post-campaign vaccination coverage increased significantly to 55.9% (95% CI=52.2%-59.6%). Multivariate logistic regression, controlled for statistically insignificant confounders of age and gender, showed that being a student HCW in 2014 (campaign year) was significantly and independently associated with influenza vaccination (OR 2.2, 95% CI=1.7-2.9, P\u3c0.001). Other significant factors were eligibility for National Immunisation Programme (NIP) funded vaccine (OR 12.3, 95% CI=6.3 – 24.0, P\u3c0.001), employment as HCWs (OR 1.9, 95% CI=1.5-2.6, P\u3c0.001), recalled campaign materials (OR 1.8, 95% CI=1.2 – 2.7, P=0.002) and enrolled in medicine (OR 1.6, 95% CI=1.1-2.4, P=0.016). Conclusions: Student HCWs’ influenza vaccination uptake improved significantly following a low-cost, peer-led promotional campaign. This approach can be adapted to other settings

    Pathways to interleukin-6 in healthy males and serious leisure male athletes: physical activity, body composition and age.

    Physical activity (PA) is beneficial to overall health, in part due to physiological changes that lower risk factors for cardiovascular disease, including reduced inflammation. However, the mechanism by which PA reduces inflammation is unclear. One possible pathway is that PA improves body composition which in turn reduces inflammation. To test this hypothesis, we used structural equation modeling (SEM) to assess PA-body composition -inflammation pathways, as well as influences of age. In a sample of 72 healthy males with a range of PA profiles (age 18-65, mean ± sd = ), we measured PA as metabolic equivalent tasks (as per the International PA Questionnaire), body composition as percent body fat, lean mass, and fat mass, and inflammation as plasma interleukin-6 (IL-6). We treated body composition in the SEM analysis as a latent variable indicated by the three measures. We performed statistical corrections for missing values and one outlier. The model demonstrated significant effects of PA on IL-6 both directly and through body composition. Percent body fat, fat mass, and lean mass were significant indicators of the body composition latent variable. Additionally, age showed an indirect effect on IL-6 through body composition, but no direct effect. The findings suggest that PA does improve inflammatory profile through improving body composition, but that other pathways also exist

    Functional Imaging of Autonomic Regulation: Methods and Key Findings.

    Central nervous system processing of autonomic function involves a network of regions throughout the brain which can be visualized and measured with neuroimaging techniques, notably functional magnetic resonance imaging (fMRI). The development of fMRI procedures has both confirmed and extended earlier findings from animal models, and human stroke and lesion studies. Assessments with fMRI can elucidate interactions between different central sites in regulating normal autonomic patterning, and demonstrate how disturbed systems can interact to produce aberrant regulation during autonomic challenges. Understanding autonomic dysfunction in various illnesses reveals mechanisms that potentially lead to interventions in the impairments. The objectives here are to: (1) describe the fMRI neuroimaging methodology for assessment of autonomic neural control, (2) outline the widespread, lateralized distribution of function in autonomic sites in the normal brain which includes structures from the neocortex through the medulla and cerebellum, (3) illustrate the importance of the time course of neural changes when coordinating responses, and how those patterns are impacted in conditions of sleep-disordered breathing, and (4) highlight opportunities for future research studies with emerging methodologies. Methodological considerations specific to autonomic testing include timing of challenges relative to the underlying fMRI signal, spatial resolution sufficient to identify autonomic brainstem nuclei, blood pressure, and blood oxygenation influences on the fMRI signal, and the sustained timing, often measured in minutes of challenge periods and recovery. Key findings include the lateralized nature of autonomic organization, which is reminiscent of asymmetric motor, sensory, and language pathways. Testing brain function during autonomic challenges demonstrate closely-integrated timing of responses in connected brain areas during autonomic challenges, and the involvement with brain regions mediating postural and motoric actions, including respiration, and cardiac output. The study of pathological processes associated with autonomic disruption shows susceptibilities of different brain structures to altered timing of neural function, notably in sleep disordered breathing, such as obstructive sleep apnea and congenital central hypoventilation syndrome. The cerebellum, in particular, serves coordination roles for vestibular stimuli and blood pressure changes, and shows both injury and substantially altered timing of responses to pressor challenges in sleep-disordered breathing conditions. The insights into central autonomic processing provided by neuroimaging have assisted understanding of such regulation, and may lead to new treatment options for conditions with disrupted autonomic function

    When it comes to mineral synthesis, there is a lot we can learn from nature. Although we can synthesize a range of materials in the laboratory, the experimental conditions are often constrained to particular ranges of temperature, pH, etc. Biological systems, on the other hand, seem to be able to produce individual minerals and complex composite mineral structures under a variety of conditions, many of which are far from those applied to create their synthetic counterparts. Understanding how nature does this could provide a means to produce novel biomimetic materials with potential applications in a diverse range of fields from medicine to materials engineering

    Nature's conveyor belt - the matrix mediated biomineralization of magnetite in chitons (Mollusca)

    Chitons are marine molluscs that use a variety of iron and calcium based minerals to harden their teeth, which they use to scrape algae growing upon, and within, rocks. The teeth are mounted on a long ribbon-like organ termed the radula, with immature, unmineralized teeth at the posterior end and the hardened iron-mineralized teeth at the anterior end (Fig. 1). At any one time, up to 80 individual tooth rows can be observed, with each row becoming progressively mineralized as it moves forward in a conveyor belt-like manner. The ability to study the entire mineralization process in a single animal makes these creatures ideal for the study of matrix mediated biomineralization. The chiton’s ability to mineralize iron has inspired researchers who believe that new biomimetic materials and technologies can be developed based on the principles of biomineral formation

    Intramuscular triglycerides (IMTG) are stored in lipid droplets (LD) in skeletal muscle, and it is thought that subcellular location, size and number of LDs relate to insulin sensitivity (IS) more so than IMTG content alone. Reduced expression and function of mitochondria has been implicated in the development of insulin resistance, leading to an accumulation of IMTG due to reduced oxidative capacity. The aim of the present study was to 1) establish a better understanding of the sex differences in the relationship between LD profile, mitochondrial density and the development of insulin resistance thereby 2) contributing to current and future research improving insulin sensitivity and preventing the progression of insulin resistance. The study investigated the hypothesis that 1) females have greater LD stores compared to males whilst maintaining insulin sensitivity, and 2) that type two diabetes (T2D) patients have greater LD stores compared to their lean and obese counterparts. Muscle biopsies were obtained from 48 male (n=24) and female (n=24) participants, categorised into groups (n=16/group) based on metabolic health; lean, obese and T2D. Cryosections (5μm) were stained using appropriate antibodies targeting myosin heavy chain I (MHC I) and MHC IIa. LDs were stained using bodipy 493/503 and mitochondria using mouse monoclonal anti-OxPhos complex IV (COXIV). Images were obtained using widefield and confocal fluorescence microscopy. In males, LD content was two-fold greater (PTII; P0.05). In conclusion, this thesis presents novel data on sex differences in LD profile, mitochondrial density and insulin resistance and suggests a potential sex paradox; whereby reduced sex hormone production in post-menopausal females, alongside inactivity maybe more important determinants of insulin sensitivity than LD profile alone

    Bank corporate governance: a proposal for the post-crisis world

    The corporate governance problems of banks are qualitatively and quantitatively different from those of other firms. The authors argue that a key factor contributing to this difference is the growing opacity and complexity of bank activities, a trend that has increased the difficulty of managing risk in financial firms. They also cite the governance challenges posed by the holding company organization of banks, in which two boards of directors—the bank’s own board and the board of the holding company that owns the bank—monitor the bank. This paradigm results in significant confusion about the role of bank holding company directors: While regulators focus on directors’ safety and soundness responsibilities, state corporate laws governing the conduct of managers focus on the conflicting goal of maximizing shareholder wealth. Reviewing the existing solutions to bank corporate governance problems, the authors argue that it is time to impose a more rigorous standard of conduct on bank directors. They contend that “post-crisis” bank directors should be held to high professional standards rather than the amateur standard that governs directors generally, and they propose “banking expert” requirements for risk committee members akin to the requirements that Sarbanes-Oxley imposes on audit committees. They further assert that all bank directors should be “banking literate,” possessing the specialized knowledge needed to monitor and control risk taking in complex banking institutions
